This Application Note is of how to make a correct upscale for depth filtration in the lab. The described filtration trials of the yeast cell suspension have shown that a linear scale-up of a depth filtration step is feasible.
This Application Note is about turbidity reduction of herbal extracts and of how to purify plant extracts using depth filtration. The aim is to retain the flavors and fragrance of the valuable herbs while filtering out all solid particles.
In a second step, the generated results of the lab scale filtration tests were used for a scale-up calculation to a batch size of 100 L for each herbal extract.
In this Application Note, the optimization of the filtration process of a mAb-contained CHO cell culture is shown in detail. So far, the suspension is filtered in three steps, before a sterile filtration using a 0.2 μm membrane is applicable. To ensure the sterile filtration step, a flocculant has to be added prior to each filtration. In order to reduce the number of process steps in the cell harvest process, filtration tests with FILTROX depth filter sheets are performed in the laboratory scale.
The aim is to determine the most suitable filter aid and its ideal quantity for the cell culture. And, in addition, to achieve turbidity values below 30 NTU to ensure subsequent sterile filtration.

Optimized solutions for Plasmid- DNA clarification and purification
PURADISC® BIO SD is the state-of-the-art technology for alluvial filtration for Plasmid-DNA clarification. It is based on depth filters that capture contaminants in their three-dimensional matrix. The used PURAFIX® depth filters are low in extractable ions and pyrogens. By adding filter aid, such as diatomaceous earth, the capacity per m2 filter area can be drastically increased.
Save one process step The main advantage of alluvial filtration for the clarification step of Plasmid-DNA is the elimination of the two-step approach. Due to the increased filtration capacity the traditional approaches such as centrifugation followed by depth filtration or a two-step depth filtration are not necessary anymore. The simultaneous reduction of impurities e.g. HCP is another advantage.

Single-use and traditional approach of clarification
The ongoing optimization of mammalian cell lines and fermentation processes leads to higher cell densities/titers and higher monoclonal antibody (mAb) yields. Due to this optimization also the biomass increases, including cell debris and other impurities, which challenges the clarification/harvest step.
The typical clarification process for mammalian cells and mAb involves a primary and a secondary clarification step. Due to potentially high shear forces and the necessity of this step to remove submicron particles, depth filtration is a mandatory step before transferring the liquid into the downstream.

An Economical Solution for Midstream Clarification
Introduction Continuous process optimization is a key factor in the biotech industry. With higher particle loads (>108 cells/ml), standard technologies for cell removal (midstream processing) – e.g. centrifugation, separation, membrane- and depth filtration – find their limits.
To increase efficiency and making the midstream process more economical alluvial filtration is the state-of-the-art solution. Alluvial filtration is a type of depth filtration and a well-established, econo-mical method in pharmaceutical industries (e.g. plasma fractionation). Instead of using an immobilized depth filter medium, filter aid (e.g. diatomaceous earth, perlite) is used to constantly build a filter cake during filtration. The filter cake with its resistance acts then as the actual filter medium. Alluvial filtration, therefore, leads to a higher filter capacity – especially with compressible particles, e.g. microbial or mammalian cells. This technology leads to a maximum product yield and highest economic efficiency and is linear scalable from lab to production scale.
